The KLOW stack combines four bioactive peptides—BPC-157, TB-500 (a synthetic fragment of thymosin beta-4), GHK-Cu (glycyl-L-histidyl-L-lysine copper complex), and KPV (lysine-proline-valine)—that have been investigated for their roles in tissue repair, inflammatory regulation, and cellular signaling. BPC-157 is a stable gastric pentadecapeptide studied for cytoprotective effects and its ability to modulate angiogenesis, nitric oxide signaling, and fibroblast activity. TB-500 has been examined for its influence on actin dynamics, cell migration, and inflammatory pathways, contributing to processes involved in tissue regeneration and wound repair. GHK-Cu is a naturally occurring copper-binding tripeptide associated with modulation of gene expression related to collagen synthesis, antioxidant defenses, and extracellular matrix remodeling. KPV, a tripeptide derived from the C-terminal sequence of α-melanocyte-stimulating hormone (α-MSH), has been investigated for its anti-inflammatory properties, including inhibition of pro-inflammatory cytokine signaling and regulation of immune cell activity. Collectively, this peptide combination has been explored in preclinical and experimental contexts for its potential effects on regenerative signaling pathways, vascular integrity, immune modulation, and structural protein turnover.
