MOTS-c (mitochondrial open reading frame of the 12S rRNA-c) is a mitochondria-derived bioactive peptide encoded by the mitochondrial genome and expressed in multiple tissues, including skeletal muscle and liver. It functions as a regulator of cellular energy homeostasis and metabolic stress responses. MOTS-c exerts its effects through modulation of nuclear gene expression and activation of intracellular signaling pathways associated with glucose utilization, fatty acid metabolism, and insulin sensitivity, including AMP-activated protein kinase (AMPK)–dependent mechanisms. Under conditions of metabolic stress, MOTS-c translocates to the nucleus, where it influences transcriptional programs involved in oxidative metabolism and cellular adaptation. In preclinical and clinical research settings, MOTS-c has been investigated for its effects on metabolic flexibility, insulin resistance, mitochondrial function, and age-associated changes in energy regulation, as well as its potential role in exercise-related and stress-responsive metabolic signaling.
